目的:观察中肺合剂对Lewis肺癌小鼠肿瘤组织血管生成的影响。方法:取C57BL/6小鼠50只,接种Lewis肺癌瘤株后,随机分阴性对照组(0.9%生理盐水),中肺合剂低、中、高剂量组(12.7、25.4、50.8g.kg-1),阳性对照组(CTX组,30mg.kg-1),每组10只。观察12天停药。无菌条件下取肿瘤组织,采用EnVision法检测肿瘤组织血管内皮生长因子(VEGF)和CD34蛋白的表达情况,并计数微血管密度(MVD)。结果:中肺合剂能降低肿瘤组织中血管内皮生长因子(VEGF)的表达和微血管密度(MVD),VEGF表达和MVD之间呈正相关(P<0.01)。其中以中、高给药剂量组抑制作用显著,与生理盐水组相比有显著差异(P<0.01)。低剂量组与生理盐水组相比无显著差异(P>0.05)。结论:中肺合剂能抑制Lewis肺癌小鼠肿瘤组织新生血管的形成,降低Lewis肺癌小鼠肿瘤组织中血管内皮生长因子(VEGF)的表达和微血管密度(MVD),以中、高剂量组最明显。
Objective: To observe the effect of Zhongfei Mixture on tumor angiogenesis in mice with Lewis lung cancer. Methods: Fifty C57BL / 6 mice were inoculated with Lewis lung cancer cell lines and randomly divided into negative control group (0.9% saline), middle and low dose group (12.7,25.4,50.8g.kg- 1), positive control group (CTX group, 30mg.kg-1), 10 rats in each group. Observed 12 days withdrawal. Tumor tissues were removed under sterile conditions. EnVision method was used to detect the expression of vascular endothelial growth factor (VEGF) and CD34 protein in tumor tissues and microvessel density (MVD) was counted. Results: Zhongfei Mixture can reduce the expression of vascular endothelial growth factor (VEGF) and microvessel density (MVD) in tumor tissue, and there was a positive correlation between VEGF expression and MVD (P <0.01). Among them, the medium and high dosage groups were significantly inhibited, compared with the saline group there was significant difference (P <0.01). There was no significant difference between the low-dose group and the saline group (P> 0.05). Conclusion: Zhongfei Mixture can inhibit the formation of neovasculature in Lewis lung cancer mice and decrease the expression of vascular endothelial growth factor (VEGF) and microvessel density (MVD) in Lewis lung cancer mice. The medium and high dose groups are the most obvious .
