LIM结构域是在展示不同生物学作用的多种蛋白质中发现的进化保守的双锌指基序。LIM蛋白可以在细胞质和细胞核中介导蛋白质与蛋白质的相互作用。Ajuba属于LIM结构域蛋白家族第三类中的Zyxin蛋白家族,其参与细胞外基质的装配,并与相关蛋白一起调节连接细胞外基质和细胞骨架的靶基因蛋白表达。并介导广泛的生物学功能,包括调控细胞周期、细胞粘附与连接、细胞迁移、细胞增殖和凋亡以及细胞分化等。文章通过对LIM蛋白家族结构,分类及其功能的介绍,从而进一步对Ajuba蛋白的发现,结构及生物学功能在癌症和糖尿病的临床应用上进行综述。
The LIM domain is an evolutionarily conserved double zinc finger motif found in a variety of proteins that demonstrate different biological roles. The LIM protein mediates protein-protein interactions in the cytoplasm and nucleus. Ajuba belongs to the family of Zyxin proteins in the third class of the LIM domain protein family, which is involved in the assembly of extracellular matrix and, together with related proteins, regulates the expression of target genes that connect extracellular matrix and cytoskeleton. And mediates a wide range of biological functions including regulation of cell cycle, cell adhesion and ligation, cell migration, cell proliferation and apoptosis, and cell differentiation. By introducing the structure, classification and function of LIM protein family, the article further reviews the clinical application of Ajuba protein discovery, structure and biological function in cancer and diabetes.
