目的探讨CCR2抑制剂RS504393对小鼠脑缺血的治疗作用。方法用野生型小鼠(C57BL/6)40只构建短暂性局限性脑缺血模型,随机分为假手术组(麻醉后,分离暴露血管,但不使用线栓堵塞大脑中动脉);模型组(给予等容积的生理盐水,腹腔注射);高剂量组(RS504393 2 mg·kg~(-1),腹腔注射);低剂量组(RS504393 1 mg·kg~(-1),腹腔注射)。使用TTC染色计算梗死体积,行为学方法评价,脑组织含水量检测等技术方法分析CCR2抑制剂RS504393对小鼠脑缺血的治疗作用。结果与模型组比较,高剂量组神经功能显著改善、脑梗死体积百分比降低、脑水肿减轻;低剂量组对该模型也有一定改善作用。结论在小鼠脑缺血早期给予CCR2抑制剂RS504393对缺血脑组织有保护作用。
Objective To investigate the therapeutic effect of CCR2 inhibitor RS504393 on cerebral ischemia in mice. Methods Forty wild type mice (C57BL / 6) were used to establish a transient focal cerebral ischemia model, which were randomly divided into sham-operation group (after anesthesia, the vessels were exposed, but the middle cerebral artery was blocked without thread); model group (Given an equal volume of saline, intraperitoneal injection); high dose group (RS504393 2 mg · kg -1, intraperitoneal injection); low dose group (RS504393 1 mg · kg -1, intraperitoneal injection). TTC staining was used to calculate the infarct volume, behavioral evaluation, brain water content detection and other technical methods to analyze the therapeutic effect of CCR2 inhibitor RS504393 on cerebral ischemia in mice. Results Compared with the model group, the neurological function of the high-dose group was significantly improved, the volume percentage of cerebral infarction was decreased, and brain edema was relieved. The low-dose group also improved the model. Conclusion The early administration of CCR2 inhibitor RS504393 in mice with cerebral ischemia has a protective effect on ischemic brain.
