目的:建立HPLC-MS法测定人血浆中埃索美拉唑含量,为药动学研究提供帮助。方法:血浆蛋白经乙腈溶液处理后,采用Zorbax SB-C18色谱柱(2.1 mm×50 mm,5μm),柱温30℃,以乙腈-0.1%甲酸为流动相,流速0.3 ml.min-1,通过电喷雾离子源,以多反应监测方式(MRM)进行离子检测,通道为m/z346.0→197.7(埃索美拉唑)结果:血浆中埃索美拉唑在5 ng.ml-1~1000 ng.ml-1浓度范围内线性关系良好,最低检测限为5 ng.ml-1。结论:该方法操作简便,分析快速,适用于人血浆中埃索美拉唑的检测。
OBJECTIVE: To establish a HPLC-MS method for the determination of esomeprazole in human plasma and to provide assistance for the study of pharmacokinetics. Methods: The plasma protein was treated with acetonitrile, and then applied on a Zorbax SB-C18 column (2.1 mm × 50 mm, 5 μm) at a column temperature of 30 ℃ with acetonitrile-0.1% formic acid as mobile phase at a flow rate of 0.3 ml · min- Ion detection was performed by electrospray ionization with multiple reaction monitoring (MRM) with a channel of m / z 346 → 197.7 (esomeprazole) Results: Plasma esomeprazole was incubated at 5 ng.ml-1 Linearity was good with the ~ 1000 ng.ml-1 concentration range, with a minimum detection limit of 5 ng.ml-1. Conclusion: The method is simple, rapid and suitable for the detection of esomeprazole in human plasma.
