目的:研究窄叶鲜卑花叶中黄酮和多酚的超声提取工艺及抗氧化活性。方法:以黄酮和多酚的总评OD值为评价指标,在单因素试验的基础上,采用响应面分析法对液料比、乙醇浓度、提取温度、提取时间等条件进行优化,并采用铁氰化钾与三氯乙酸体系、1,1-二苯基-2-三硝基苯肼(DPPH·)法研究其抗氧化性。结果:最佳工艺条件为:浸泡时间60 min,液料比62.2∶1(m L/g),乙醇浓度63.7%,提取温度61℃,超声(250 W)提取时间67.2 min,黄酮和多酚的提取率分别为140.28和224.54 mg/g。0.80 mg/m L窄叶鲜卑花叶提取液与0.02 mg/m L抗坏血酸对DPPH·自由基清除能力相当,清除率为82.0%。结论:该响应面法优化得到的提取工艺稳定合理,窄叶鲜卑花叶具有较好的抗氧化能力。
OBJECTIVE: To study the ultrasonic extraction process and antioxidant activity of flavonoids and polyphenols in narrow-leaved Xianbei mosaic leaves. Methods: Based on the single factor test, the total OD value of flavonoids and polyphenols was used as evaluation index to optimize the conditions of liquid to material ratio, ethanol concentration, extraction temperature and extraction time by using response surface methodology. Potassium and trichloroacetic acid system, 1,1-diphenyl-2-trinitrophenylhydrazine (DPPH ·) method to study its antioxidant activity. Results: The optimum conditions were as follows: the soaking time was 60 min, the ratio of liquid to material was 62.2:1 (m L / g), the concentration of ethanol was 63.7%, the extraction temperature was 61 ℃, the extraction time was 250 min, the extraction time was 67.2 min, The extraction rates were 140.28 and 224.54 mg / g, respectively. 0.80 mg / m L narrow-leaf Xianbei mosaic extract and 0.02 mg / m L ascorbic acid on DPPH · free radical scavenging capacity, clearance rate of 82.0%. Conclusion: The extraction process optimized by response surface methodology is stable and reasonable, and the narrow-leaf Xianbei mosaic has good antioxidant capacity.
