目的建立原发瘤的神经母细胞瘤体外细胞系,制作荷瘤鼠模型。方法术中取新鲜肿瘤标本,利用酶消化法,建立起体外细胞系,接种于裸鼠(N=11)和SCID小鼠(N=3)后肢前下方的皮下及腹膜后,接种2.5×107/ml的单细胞悬液0.5ml。结果人神经母细胞瘤原发瘤体外细胞系建立成功,现已传至第52代。裸鼠(5/11)和SCID小鼠(N=2/3)荷瘤成功,全身转移有明显恶液质体征的裸鼠2只,其余为接种局部肿瘤生长。结论建立神经母细胞瘤细胞系对于研究其转基因诱导分化、神经母细胞瘤的分子生物学特征及其肿瘤自消机制打下了良好的基础。
Objective To establish a primary tumor-bearing neuroblastoma cell line in vitro and make a tumor-bearing mouse model. Methods Fresh tumor samples were collected during operation. In vitro cell lines were established by enzymatic digestion and inoculated subcutaneously and retroperitoneally into nude mice (N = 11) and SCID mice (N = 3) / ml of single cell suspension 0.5ml. Results Human neuroblastoma primary tumor cell line established in vitro, has now reached the 52nd generation. Nude mice (5/11) and SCID mice (N = 2/3) were successfully tumor-bearing. Two nude mice with systemic signs of malignant liquefaction were transplanted into the body. The rest were local tumor growth. Conclusion Establishment of neuroblastoma cell lines for the study of its transgenic induced differentiation, neuroblastoma molecular biology and tumor self-elimination mechanism has laid a good foundation.
