目前多数学者认为,LAK细胞本质上是NK细胞,在IL-2诱导下其细胞毒活性增强,为LAK现象。在LAK细胞研究中,关于IL-2诱导下LAK活性增高与细胞增殖之间的关系,文献报道互有矛盾。鉴于HNK1抗原选择性地表达于NK和K细胞,LAK细胞HNK1亦为刚性,本实验从IL-2诱导前后HNK1阳性率的变化及LAK功能的流式细胞仪分析,探讨了LAK细胞活性与细胞增殖的关系。结果表明,在本实验条件下,IL-2可诱导出LAK活性,但HNK1阳性细胞数不增加,提示LAK细胞活性可能与细胞增殖无关。实验中还观察到去单核细胞使IL-2诱导的LAK活性下降,提示单核细胞在IL-2诱导下也有LAK活性。
At present, most scholars believe that LAK cells are NK cells in nature, and their cytotoxicity is enhanced by IL-2, which is LAK phenomenon. In the study of LAK cells, the relationship between the increase of LAK activity and cell proliferation induced by IL-2 has been contradictory in the literature. In view of HNK1 antigen selective expression in NK and K cells, LAK HNK1 also rigid, in this experiment from before and after IL-2 induction of HNK1 positive rate changes and LAK function of flow cytometry analysis of LAK cell activity and cells Proliferation relationship. The results showed that under the experimental conditions, IL-2 induced LAK activity, but the number of HNK1-positive cells did not increase, suggesting that LAK cell activity may not be related to cell proliferation. It was also observed in experiments that IL-2-induced LAK activity was decreased in monocytes, suggesting that monocytes also have LAK activity induced by IL-2.
