目的初步探讨大肠癌中聚(腺苷二磷酸核糖)水解酶[Poly(ADP-ribose)glycohydrolase,PARG]与聚(腺苷二磷酸核糖)聚合酶[Poly(ADP-ribose)polymerase,PARP]的关系。方法应用S-P免疫组化检测人大肠癌组织中PARG、PARP的表达;以PARG抑制剂丹宁酸(Gallotannin,GLTN)为处理因素,采用流式细胞计数检测小鼠结肠腺癌CT26细胞中PARG、PARP表达变化。结果PARG和PARP在人大肠癌组织中的表达呈正相关(r=0.300 01,P<0.05);流式细胞计数显示,用GLTN处理后,小鼠结肠腺癌CT26细胞中PARG、PARP的阳性细胞率分别较对照组降低,差异具有统计学意义(P<0.05)。结论大肠癌中PARG和PARP的表达有相关性,PARG抑制剂可以抑制PARG及PARP表达,PARG可能对PARP具有调节作用。
Objective To investigate the expression of poly (ADP-ribose) polymerase (PARP) and poly (ADP-ribose) polymerase (PARP) relationship. Methods SP immunohistochemical method was used to detect the expression of PARG and PARP in colorectal cancer tissues. PARG inhibitor GLN was used as the treatment factor. Flow cytometry was used to detect the expression of PARG, PARP expression changes. Results The expression of PARG and PARP in colorectal carcinoma was positively correlated (r = 0.300 01, P <0.05). Flow cytometry showed that PARG and PARP positive cells in colon adenocarcinoma CT26 cells treated with GLTN The rates were lower than the control group, the difference was statistically significant (P <0.05). Conclusion The expressions of PARG and PARP in colorectal cancer are correlated. PARG inhibitor can inhibit the expression of PARG and PARP, and PARG may play a regulatory role in PARP.
