目的 探讨顺铂对耳蜗单离听觉细胞诱导型─氧化氮合酶(iNOS/NOSⅡ)表达的影响。方法 以单 离毛细胞和单离螺旋神经节神经元为模型,用免疫组化方法(ABC法)研究顺铂对细胞内NOSⅡ表达的影响。结果 在1mM顺铂平衡盐溶液中室温下培养两小时的外毛细胞及培养半小时的单离螺旋神经元与对照组相比显示了 对NOSⅡ抗体较强的免疫反应。结论 提示顺铂诱导了单离外毛细胞及单离螺旋神经元内NOSⅡ的合成增加,NO 可能参与了顺铂的耳毒作用。
Objective To investigate the effect of cisplatin on the expression of inducible nitric oxide synthase (iNOS / NOSⅡ) in isolated cochlear auditory cells. Methods The isolated mononuclear cells and isolated ganglion neurons were used as a model to study the effect of cisplatin on the expression of intracellular NOSⅡ by immunohistochemistry (ABC method). Results The immunoreactivity against NOS II antibody was shown to be higher in the 1 mM cisplatin balanced salt solution at room temperature for two hours compared to the control group when cultured for two hours in outer hair cells and cultured for half an hour in isolated spiral neurons. Conclusions Cisplatin induced an increase in the synthesis of NOSII in single isolated outer hair cells and isolated spheroidal neurons, and NO may be involved in cisplatin ototoxicity.
