目的:建立反相高效液相色谱法同时测定不同根龄虎杖根中虎杖苷和白藜芦醇含量。方法:采用Kromasil C18(4.6 mm×250 mm,5μm)色谱柱,流动相甲醇-1.0%磷酸水溶液(50∶50),流速0.8 mL.min-1,检测波长319 nm,柱温为30℃。结果:虎杖苷和白藜芦醇的进样量分别在0.716~7.160μg(r=0.999 6)和0.740~7.400μg(r=0.999 8)与峰面积呈良好的线性关系;平均回收率(n=6)分别为99.7%,98.5%。结论:本方法快速简便,重复性好,专属性强,可作为虎杖根的质量控制方法。
Objective: To establish an RP-HPLC method for the simultaneous determination of polydatin and resveratrol in roots of Polygonum cuspidatum roots. Methods: Kromasil C18 column (4.6 mm × 250 mm, 5 μm) was used. The mobile phase consisted of methanol-1.0% phosphoric acid (50:50) and the flow rate was 0.8 mL.min-1. The detection wavelength was 319 nm and the column temperature was 30 ℃. Results: The injection volume of polydatin and resveratrol showed a good linear relationship with the peak area in the range of 0.716 ~ 7.160μg (r = 0.999 6) and 0.740 ~ 7.400μg (r = 0.999 8). The average recovery (n = 6) were 99.7%, 98.5% respectively. Conclusion: The method is rapid, simple, reproducible and specific, and can be used as quality control method for Polygonum cuspidatum.
