目的:探讨柴黄胃溃宁对肝郁脾虚证胃溃疡大鼠脑组织中HSP70 mRNA和Caspase-3 mRNA基因表达的影响和可能的疗效机制。方法:采用多因素复合模拟中医病因结合乙酸法建立肝郁脾虚证胃溃疡大鼠模型,用HE染色法观察大鼠胃窦部溃疡组织的病理学改变,采用RT-PCR技术检测脑组织中HSP70mRNA、Caspase-3mRNA表达水平。结果:模型组大鼠脑组织中HSP70mRNA、Caspase-3mRNA表达明显升高,柴黄胃溃宁中剂量组和法莫替丁组HSP70mRNA的表达水平升高,Caspase-3mRNA的表达水平降低,而柴黄胃溃宁高剂量组结果相反。结论:柴黄胃溃宁对肝郁脾虚证胃溃疡模型大鼠神经系统有显著的调节作用,以中剂量效果最佳。
Objective: To investigate the effect of Chaihuang Weicuning on gene expression of HSP70 mRNA and Caspase-3 mRNA in brain tissue of gastric ulcer rats with liver-qi stagnation and spleen-deficiency syndrome and its possible mechanism. Methods: The model of gastric ulcer in rats with liver-qi stagnation and spleen deficiency syndrome was established by multivariate analysis to simulate the etiology of TCM and acetic acid method. The pathological changes of gastric ulcer in rats were observed by HE staining. The expression of HSP70mRNA , Caspase-3mRNA expression level. Results: The expression of HSP70mRNA and Caspase-3mRNA in the model group were significantly increased, the expression of HSP70mRNA in Casimuening Weining middle dosage group and famotidine group were increased, the expression of Caspase-3mRNA was decreased, Huang Guikui Ning high-dose group the contrary. Conclusion: Chaihuang Weicuning can significantly regulate the nervous system of rats with gastric ulcer caused by liver-qi and spleen-qi syndrome, with the best effect in middle dose.
