目的研究川芎嗪对心肌缺血大鼠模型及心肌细胞凋亡的影响。方法建立大鼠心肌缺血模型,将24只大鼠按随机化原则分为对照组、高剂量组、低剂量组、假手术组。各组均测定心肌缺血和梗死范围以及CPK、LDH指标,并应用脱氧核糖核苷酸末端转移酶介导的缺口末端标记法(TUNEL法)测定各组心肌细胞凋亡及凋亡指数,比较各组间差异。结果高剂量组较对照组和低剂量组能明显降低心肌缺血大鼠模型的危险指数,降低心肌中CPK、LDH的升高。高剂量组能明显降低心肌细胞的凋亡指数,对心肌细胞凋亡的保护作用均明显高于生理盐水和低剂量组,与假手术组无统计学差异。结论川芎嗪对心肌缺血大鼠模型心肌梗死有明显保护作用,其作用机制可能与抗心肌细胞凋亡作用有关。
Objective To study the effects of tetramethylpyrazine on rat model of myocardial ischemia and cardiomyocyte apoptosis. Methods A rat model of myocardial ischemia was established. Twenty-four rats were divided into control group, high-dose group, low-dose group and sham-operation group according to the principle of randomization. Myocardial ischemia and infarct size and CPK and LDH were measured in each group. Cardiomyocyte apoptosis and apoptosis index were determined by deoxyribonucleotide terminal transferase-mediated nick end labeling (TUNEL) method. Differences between groups. Results Compared with the control and low dose groups, the high-dose group significantly reduced the risk index of the rat model of myocardial ischemia and reduced the increase of CPK and LDH in the myocardium. High-dose group can significantly reduce the apoptotic index of cardiomyocytes. The protective effect on cardiomyocyte apoptosis was significantly higher than that of saline and low-dose groups, and there was no statistical difference between the high-dose group and the sham operation group. Conclusion Ligustrazine has a protective effect on myocardial infarction in rat model of myocardial ischemia, and its mechanism may be related to the anti-apoptosis effect of cardiomyocytes.
