南方红豆杉悬浮培养细胞团区域化及营养功能分化的研究

对南方红豆杉 Taxus chinensis var.mairei细胞悬浮培养过程中的分化、细胞团区域化及营养功能分化等现象进行了研究。通过光学显微镜和电子显微镜对细胞团切片及淀粉颗粒染色从形态学上进行系统观察 ,发现悬浮培养细胞团从外向里呈明显的 3个区域 :表层细胞由成年细胞组成 ,中层细胞是由增殖能力旺盛的细胞组成 ,中心区域的细胞细胞核已消失 ,出现分化的管状分子。在电镜下可见胞间连丝。通过对细胞内淀粉含量及其代谢、可溶性蛋白含量、细胞生物量和总紫杉醇产量的测定结果 ,可以初步判断 3部分区域各自承担着不同的功能 ,细胞团是功能化的细胞结合体 The differentiation of Taxus chinensis var. mairei cells during cell suspension culture, cell cluster regionalization, and nutrient function differentiation were studied. The morphological observation of cell pellets and starch granules was carried out by light microscopy and electron microscopy. It was found that the suspension cultured cells showed three distinct regions from the outside to the outside: the superficial cells consisted of adult cells, and the middle cells were proliferated. The vigorous cell composition, the central cell nucleus has disappeared, and differentiated tubular molecules appear. Intercelluar filaments can be seen under the electron microscope. Through the determination of intracellular starch content and its metabolism, soluble protein content, cell biomass and total paclitaxel yield, it can be preliminarily judged that each of the three regions bears different functions, and the cell mass is a functional cell conjugate.
