目的:通过急性毒性实验确定顺铂对秀丽隐杆线虫24h的半数致死浓度(median lethal concentration,LC50),然后根据LC50确定亚致死暴露浓度,为进一步的亚急性毒性实验提供参考依据。方法用K-medium溶液配制不同浓度的顺铂样品溶液,K-medium溶液作对照液,同步化后的秀丽隐杆线虫分别暴露于样品液和对照液,观察24h后秀丽隐杆线虫的死亡与存活情况,同时观察24h线虫的生长发育、繁殖和运动情况,用Probit方法计算秀丽隐杆线虫的24h LC50。结果顺铂对秀丽隐杆线虫24h LC50为617 mg?L-1,95%置信区间为580~647 mg?L-1;在实验过程可发现线虫的生长发育、繁殖和运动能力都受到抑制,随着顺铂浓度的升高,抑制越明显。结论顺铂对秀丽隐杆线虫具有一定的毒性,特别是对线虫的生长发育、运动和繁殖能力都有不同程度的影响。“,”Objective To determine the median lethal concentration (LC50)of cisplatin to Caenorhabditis elegans in the acute toxicity test of 24h,Then according to LC50 to determine the sublethal exposure concentration, which provides reference for further subacute toxicity experiments. Methods K-medium solution was used to prepare a series of concentrations of cisplatin as the experimental groups, K-medium solution as the control solution. Synchronized Caenorhabditis elegans exposure in experimental and control solutions 24h,then observe the mortality, growth, development,reproduction and movement of C. elegans. LC50 were derived through Probits analysis. Results The LC50 was 617mg.L-1, and its 95%confidence interval was 580~647 mg.L-1, the growth and development, reproduction and movement ability of the nematodes were inhibited in the experimental process. Increasing the concentration of cisplatin, the restrain more obvious. Conclusion Cisplatin has a certain degree of toxicity to the nematodes, especially,the effects on the growth and development, movement and reproduction ability of the nematode is different.
