目的:探讨成年和老龄大鼠外周和中枢内催产素和加压素的含量变化。方法:采用健康成年SpragueDawley(SD)雄性大鼠和一年半的老龄化雄性SD大鼠各4只,应用免疫组织化学方法对下丘脑视上核(SON)及室旁核(PVN)中催产素(OXT)及加压素(VP)进行染色研究;同时应用ELISA方法检测两组动物血浆中的激素浓度水平。结果:与成年大鼠相比,老龄大鼠下丘脑视上核和室旁核中的OXT及VP神经元数量变化不明显,但其免疫反应染色强度显著降低,灰度分析显示单个神经元和整个核团的光密度值明显下降(P<0.05);正中隆起处VP和OXT纤维终末及膨体的免疫反应性也明显减弱。与此相对,Elisa检测显示老龄鼠血浆中的VP和OXT水平高于正常大鼠,且以VP的升高更为明显。结论:老龄鼠外周血和中枢神经元中的OXT、VP含量水平呈相反变化趋势,这可能与老龄鼠中的激素释放增加、储存减少有关,而这一变化可能参与某些老年相关疾病如水盐代谢障碍、高血压、学习记忆障碍的发生发展过程。
Objective: To investigate the changes of oxytocin and vasopressin in the peripheral and central adult rats. Methods: Four adult Sprague Dawley (SD) male rats and one and a half aged male Sprague-Dawley (SD) male rats were used in each study. Immunohistochemistry was used to detect oxytocin in the supraoptic nucleus (SON) and paraventricular nucleus (PVN) (OXT) and vasopressin (VP). Meanwhile, the levels of plasma hormone in two groups of animals were detected by ELISA. Results Compared with adult rats, the number of OXT and VP neurons in the hypothalamus supraoptic nucleus and paraventricular nucleus of aged rats was not significantly changed, but the intensity of immunoreactive staining was significantly decreased. The gray scale analysis showed that single neurons and whole The optical density of the nucleus decreased significantly (P <0.05). The immunoreactivity of VP and OXT fibers in the middle of the bulge was also significantly weakened. In contrast, the Elisa test showed that the plasma levels of VP and OXT in old rats were higher than those in normal rats, and the increase of VP was more obvious. CONCLUSION: OXT and VP contents in peripheral blood and CNS neurons in aged rats show the opposite trend, which may be related to the increase of hormone release and storage reduction in aged rats, and this change may be involved in some elderly-related diseases such as water and salt Metabolic disorders, hypertension, learning and memory disorders occurred in the development process.
