目的探讨三氧化二砷(As2O3)对A375人恶性黑素瘤细胞株酪氨酸酶活性及黑素生成的影响。方法利用四甲基偶氮唑蓝(MTT)还原法测定细胞活力,采用酶学方法测定酪氨酸酶活性,465nm比色法测定黑素含量。结果As2O3浓度<0.01μmol/L时,随As2O3浓度的增加,促进黑素瘤细胞的增殖、黑素的合成及上调酪氨酸酶的活性;As2O3浓度等于0.01μmol/L时作用最强。0.1~1μmol/L则表现为抑制作用;>1μmol/L则出现细胞毒性作用。结论经As2O3处理后A375人恶性黑素瘤细胞株的细胞增殖、酪氨酸酶活性及黑素的生成与As2O3浓度呈双相性变化。
Objective To investigate the effects of arsenic trioxide (As2O3) on the tyrosinase activity and melanogenesis of A375 human malignant melanoma cell line. Methods MTT assay was used to determine cell viability. The tyrosinase activity was determined by enzymatic method. The content of melanin was determined by 465nm colorimetric assay. Results When As2O3 concentration was less than 0.01μmol / L, the proliferation of melanoma cells, melanin synthesis and tyrosinase activity were increased with the increase of As2O3 concentration. When As2O3 concentration was equal to 0.01μmol / L, the effect was the strongest. 0.1 ~ 1μmol / L showed inhibition;> 1μmol / L cytotoxicity. Conclusion The cell proliferation, tyrosinase activity and the formation of melanin in As2O3 treated A375 human malignant melanoma cell line have biphasic changes with As2O3 concentration.
