目的 利用抗CD分子单抗,对多肽在体外激活T淋巴细胞后,进行识别计数,确定其有无生物活性。方法 用脾细胞培养物中加入多肽激活脾细胞,以不同抗CD分子单抗作标记,用流式仪检测计数。结果 证实多肽中有组分,能直接激活和分化T淋巴细胞,并保持CD4/CD8比值恒定,在25ug-50ug/ml之间能明显提升CD4+、CD8+细胞的数量,有统计学意义。结论 特别是CD4+和CD8+细胞的增加,对中止病毒扩散和清除感染细胞是有利的,为重建艾滋病患者免疫提供新的途径。
OBJECTIVE: To use anti-CD monoclonal antibody to identify and count T lymphocytes in vitro and determine whether it has biological activity. Methods Spleen cells were cultured with splenocytes to activate splenocytes and labeled with different anti-CD monoclonal antibodies. Flow cytometry was used to detect the counts. The results confirmed that there are components in the polypeptide that can directly activate and differentiate T lymphocytes and keep the ratio of CD4 / CD8 constant. The number of CD4 + and CD8 + cells can be obviously increased between 25ug-50ug / ml with statistical significance. Conclusion The increase of CD4 + and CD8 + cells, in particular, is beneficial to stop the spread of virus and eliminate infected cells, and provide a new way to rebuild AIDS patients’ immunity.
