制备新生2d龄大鼠纹状体提取液及其不同分子量的组分,用快速自动比色微量分析法检测它们对体外培养的14d龄大鼠胚胎中脑黑质神经元的影响。结果表明,纹状体提取液有支持中脑黑质神经元生存及增强其活性的作用,而且具有剂量依赖关系,其最适浓度为1.25μg/ml。纹状体提取液还能促进中脑黑质神经元对2H-DA和3H-GABA的摄取,同对照组相比,有显著性差异(P<0.01)。通过检测纹状体提取液小于10kD、10~30kD和大于30kD3种不同分子量组分对中脑黑质神经元的作用,证明有活性的部分是分子量为10~30kD的组分。
The 2-day-old rat striatum extract and its components with different molecular weights were prepared and their effects on the substantia nigra neurons in midbrain of 14-day-old rat embryos cultured in vitro were detected by rapid automated colorimetric microanalysis. The results showed that striatal extract had the effect of supporting the survival and enhancing the activity of substantia nigra neurons in midbrain, and had a dose-dependent relationship with the optimal concentration of 1.25μg / ml. Striatal extract can also promote the uptake of 2H-DA and 3H-GABA into midbrain substantia nigra neurons, compared with the control group, there was a significant difference (P <0.01). By detecting the role of striatum extract of less than 10kD, 10 ~ 30kD and greater than 30kD three kinds of different molecular weight components of the brain cells in the substantia nigra neurons, the active part of the molecular weight of 10 ~ 30kD components.
