目的 探讨全脑缺血再灌注后大鼠脑神经细胞中NF-κB的表达及其作用。方法 28只Sprague Dawley大鼠,随机分成假手术组(对照组)以及复苏后3、6、12、24h组。采用窒息合并0.5mmol/L冰氯化钾停跳液致大鼠心跳骤停-心肺复苏模型,停跳5min后开始心肺复苏,采用免疫组化法观察复苏后各时间段神经细胞中NF-κB的表达情况。结果 复苏后6、12h海马区NF-κB表达明显增加,至24h已达11.2%,与对照组比较均有显著差异(P<0.05);海马区周围的胶质细胞也有NF-κB表达。结论 NF-κB在大鼠全脑缺血再灌注后神经细胞中的表达增加,可能具有保护和损伤双重作用。
Objective To investigate the expression of NF-κB in rat brain after global cerebral ischemia-reperfusion and its role. Methods Twenty-eight Sprague Dawley rats were randomly divided into sham operation group (control group) and 3,6,12,24 h after resuscitation. Cardiac arrest was induced by asphyxial injection of 0.5 mmol / L ice-cold potassium chloride cardioplegia in cardiopulmonary resuscitation (CPR) model. Cardiopulmonary resuscitation (CPR) was initiated 5 minutes after cardioplegia. Immunohistochemistry was used to observe the changes of NF-κB The expression of the situation. Results The expression of NF-κB in hippocampus increased significantly at 6 and 12 h after resuscitation, reaching 11.2% at 24 h after resuscitation, which was significantly different from that in control group (P <0.05). The expression of NF-κB was also observed in glial cells around hippocampus. Conclusion NF-κB may increase the expression of NF-κB in neurons after global cerebral ischemia-reperfusion in rats, which may play a dual role in protection and injury.
