目的:建立以高效液相色谱法测定锁阳中熊果酸含量的方法,检测不同产地锁阳中熊果酸的含量。方法:色谱柱为Zorbax SBC18(150mm×4.6mm,5μm);流动相为甲醇-0.085%磷酸溶液(90∶10);检测波长220nm,流速1.0mL.min-1。结果:熊果酸在0.03~0.48mg·mL-1范围内与峰面积积分值呈良好的线性关系(r=0.9996,n=5)。不同产地锁阳中熊果酸的含量在0.038%~0.445%之间。结论:不同产地锁阳中熊果酸含量差异很大。本方法简便、准确、重复性好,可用于锁阳的质量控制。
Objective: To establish a method for determination of ursolic acid in Cynomorium by high performance liquid chromatography (HPLC) and detect the contents of ursolic acid in Cynomorium of different origins. METHODS: The chromatographic column was Zorbax SBC18 (150mm×4.6mm, 5μm); the mobile phase was methanol-0.085% phosphoric acid solution (90:10); the detection wavelength was 220nm and the flow rate was 1.0mL.min-1. RESULTS: There was a good linear relationship between ursolic acid and the integrated value of peak area in the range of 0.03-0.48 mg·mL-1 (r=0.9996, n=5). The contents of ursolic acid in Cynomorium of different origins were between 0.038% and 0.445%. Conclusion: The content of ursolic acid in Cynomorium in different areas varies greatly. The method is simple, accurate, and reproducible, and can be used for quality control of Cynomorium.
