目的观察原花青素B2(proanthocyanidin-B2,PC-B2)对体内外蛋白质非酶糖基化终产物(AGEs)形成的影响。方法用链脲佐菌素建立大鼠糖尿病模型,ig不同剂量(25、50、100 mg/kg)PC-B2 12周,检测血糖和AGEs的量。同时将不同质量浓度PC-B2分别与葡萄糖和牛血清白蛋白、半乳糖和牛血清白蛋白在37℃孵育60 d,用荧光光度法测定AGEs的荧光值。结果 PC-B2能明显降低糖尿病大鼠血糖(P<0.01),抑制AGEs的形成(P<0.05),并呈明显的剂量依赖性。PC-B2对体外AGEs的生成也有明显的抑制作用(P<0.01)。结论 PC-B2可明显降低糖尿病大鼠血糖,对体内外AGEs的形成具有明显的抑制作用。
Objective To observe the effect of proanthocyanidin-B2 (PC-B2) on the formation of non-enzymatic glycation end products (AGEs) in vitro and in vivo. Methods Diabetic rats were induced with streptozotocin (STZ). The PC-B2 at different doses (25, 50 and 100 mg / kg) were given for 12 weeks. The blood glucose and AGEs were measured. At the same time, different concentrations of PC-B2 were respectively incubated with glucose, bovine serum albumin, galactose and bovine serum albumin for 60 days at 37 ℃. The fluorescence of AGEs was measured by fluorescence spectrophotometry. Results PC-B2 significantly decreased the blood glucose (P <0.01), inhibited the formation of AGEs (P <0.05), and showed a dose-dependent manner. PC-B2 also inhibited AGEs production in vitro (P <0.01). Conclusion PC-B2 can significantly reduce the blood glucose in diabetic rats, and has a significant inhibitory effect on the formation of AGEs in vitro and in vivo.
