目的探讨淫羊藿苷对异丙肾上腺素诱导H9C2大鼠心肌细胞心肌重塑模型CYP27B与维生素D受体mRNA表达的影响。方法将H9C2细胞随机分为6个组:对照组,模型组,淫羊藿苷0.1μmol/L+模型组,淫羊藿苷1μmol/L+模型组,淫羊藿苷10μmol/L+模型组,淫羊藿苷100μmol/L+模型组,利用浓度为80μmol/L的异丙肾上腺素(ISO)进行造模。MTT法观察H9C2心肌细胞增殖活性,Real-Time QPCR法检测CYP27B与维生素D受体mRNA表达。结果 MTT结果显示与对照组相比模型组细胞存活率下降(P<0.05),经过淫羊藿苷处理后细胞存活率提高(P<0.05)。Real-TimeQPCR显示模型组CYP27B与维生素D受体基因mRNA表达量相对对照组均降低(P<0.05),不同浓度淫羊藿苷作用后CYP27B与VDR基因表达量相对模型组则上升(P<0.05)。结论淫羊藿苷可以通过调节维生素D轴对异丙肾上腺素诱导的H9C2细胞心肌重塑模型产生保护作用。
Objective To investigate the effects of icariin on the expression of CYP27B and vitamin D receptor mRNA in myocardial cells remodeling induced by isoprenaline in H9C2 rat cardiac myocytes. Methods H9C2 cells were randomly divided into 6 groups: control group, model group, icariin 0.1μmol / L + model group, icariin 1μmol / L + model group, icariin 10μmol / L + model group, The model group was made of 100gol / L and 50gol / L isoproterenol (ISO). MTT assay H9C2 cardiomyocyte proliferation activity, Real-Time QPCR assay CYP27B and vitamin D receptor mRNA expression. Results MTT results showed that compared with the control group, the cell viability of the model group decreased (P <0.05), and the cell survival rate increased after icariin treatment (P <0.05). Compared with the control group, the mRNA expression of CYP27B and vitamin D receptor in Real-TimeQPCR model group was lower than that in the control group (P <0.05), while the expression of CYP27B and VDR gene in model group increased after icariin treatment ). Conclusion Icariin can protect isoproterenol-induced myocardial remodeling in H9C2 cells through regulating the vitamin D axis.
