目的:观察羊膜移植对准分子激光屈光性角膜切削术(photorefractive keratectomy,PRK)后角膜组织表达转移生长因子-β_1(transforming growth factor-β_1,TGF-β_1)、Ⅰ、Ⅲ型胶原和纤维连接蛋白(fibronectin,FN)的影响,探讨羊膜移植减轻瘢痕形成的部分机制。方法:对10只新西兰白兔双眼行PRK,术后1眼立即行角膜表面羊膜移植术,另眼作为对照。于术后4周进行免疫组织化学染色检查。结果:羊膜移植组角膜上皮和基质中角膜基质细胞及胶原纤维TGF-β_1表达较对照组明显减弱,前角膜基质表达Ⅲ型胶原和FN亦明显较对照组减弱。角膜基质Ⅰ型胶原表达两组间无明显差异。结论:羊膜能抑制PRK后角膜组织TGF-β_1、Ⅲ型胶原和FN的表达,羊膜移植后瘢痕形成轻可能部分与羊膜调节组织TGF-β_1、Ⅲ型胶原和FN的表达有关。眼科学报2000;16:239~242。
OBJECTIVE: To observe the effect of amniotic membrane transplantation on the expression of transforming growth factor-β 1 (TGF-β_1), collagen type Ⅰ and Ⅲ and fibronectin in corneal tissue after photorefractive keratectomy (PRK) (Fibronectin, FN) of the impact of amniotic membrane transplantation to reduce scar formation part of the mechanism. Methods: PRK was performed in 10 New Zealand white rabbits. Corneal surface amniotic membrane transplantation was performed in 1 eye and the other eye was used as a control. Four weeks after operation, immunohistochemical staining was performed. Results: The expression of TGF-β 1 in corneal stroma cells and collagen fibers in corneal epithelium and stroma in amniotic membrane transplantation group was significantly lower than that in control group. The expression of type Ⅲ collagen and FN in anterior corneal stroma was also weaker than that in control group. Corneal stroma type I collagen expression between the two groups no significant difference. CONCLUSION: Amniotic membrane can inhibit the expression of TGF-β 1, type Ⅲ collagen and FN in corneal tissue after PRK. The slight formation of scar may be related to the expression of TGF-β 1, type Ⅲ collagen and FN in amniotic membrane after amniotic membrane transplantation.
