目的 了解肾综合征出血热病毒云南地区滇绒鼠分离株E - 14 2的分子生物学特征。方法 应用PCR方法克隆了该株全S基因。结果 序列测定结果表明E - 14 2株的S片段的全基因序列共 170 2个核苷酸 ,编码 4 2 9个氨基酸。四种核苷酸的比例分别为A32 79% ,G2 5 12 % ,T2 3 0 2 % ,C19 0 7% ,GC含量为 4 4 19% ,AT含量为5 5 81%。E - 14 2与汉坦病毒S基因核苷酸和氨基酸同源性比较表明 ,E - 14 2株与HTN型同源率为 86 6 %~90 4 % ,而与SEO型仅为 6 8 3%~ 6 9 6 % ,E - 14 2株病毒属于HTN型 ,但与HTN型其它毒株相差高达 9 6 %~13 4 %。结论 E - 14 2株病毒是HTN型的新亚型毒株。
Objective To understand the molecular biological characteristics of E - 14 2 isolate from Yunnan hetmania hemorrhagic fever virus (HSV). Methods The whole S gene was cloned by PCR. Results The sequencing results showed that the whole S sequence of E - 14 2 strain was 170 2 nucleotides in length and encoded 429 amino acids. The proportions of the four nucleotides were A32 79%, G2 5 12%, T2 302%, C19 0 7%, GC content 4 4 19% and AT content 5 5 81%, respectively. Comparison of the nucleotide and amino acid homologies between the E - 14 2 gene and the S gene of Hantavirus showed that the homology between E - 14 2 and HTN was 86 6% ~ 90 4%, while it was only 683% with SEO % ~ 6 9 6%. E - 14 2 viruses belonged to HTN, but they differed from other HTN strains by 96% ~ 134%. Conclusion E - 14 2 viruses are new subtype of HTN.
