目的:探讨活化的p38MAPK在苦参碱(Matrine,Mat)诱导人Burkitt′s淋巴瘤Raji细胞凋亡过程中对Fas/FasL的影响。方法:选用Raji细胞进行体外培养,用苦参碱(终浓度为0.4、0.8、1.6 mg/mL)和在此基础上加入SB203580(p38 MAPK抑制剂)作用48 h后,分别采用AnnexinⅤ-FITC/PI双染法检测细胞凋亡率和Western blot检测P-p38MAPK、Fas、FasL蛋白表达的变化。结果:终浓度为0.4、0.8、1.6 mg/mL苦参碱对Raji细胞总凋亡率显著高于SB203580组及对照组(P<0.05或P<0.01);Western blot显示P-p38MAPK、Fas、FasL蛋白表达水平随苦参碱浓度的提高而增加,并且在加入SB203580后降低。P-p38MAPK与Fas、FasL具有明显相关性。结论:苦参碱可能通过活化p38MAPK来上调Fas、FasL的表达,从而促进Raji细胞凋亡。
AIM: To investigate the effect of activated p38MAPK on Fas / FasL in the process of Raji cell apoptosis induced by matrine (Mat) in human Burkitt’s lymphoma. Methods: Raji cells were cultured in vitro and treated with matrine (0.4, 0.8, 1.6 mg / mL) and SB203580 (p38 MAPK inhibitor) for 48 h. The cells were stained with AnnexinⅤ-FITC / PI double staining was used to detect the apoptotic rate and Western blot was used to detect the expression of P-p38MAPK, Fas and FasL. Results: The total apoptosis rate of Raji cells at the concentrations of 0.4, 0.8 and 1.6 mg / mL was significantly higher than that of SB203580 group and the control group (P <0.05 or P <0.01); Western blot showed that the expressions of P-p38MAPK, Fas, FasL protein expression increased with the increase of matrine concentration, and decreased after addition of SB203580. P-p38MAPK and Fas, FasL has a significant correlation. Conclusion: Matrine may promote the apoptosis of Raji cells by up-regulating the expression of Fas and FasL by activating p38MAPK.
