目的综合评价CYP17基因多态性与中国女性乳腺癌遗传易感性的关联强度。方法通过检索万方,维普,CNKI,Medline,springerlink和PUBMED等中英文数据库,获取CYP17基因多态性与中国女性乳腺癌遗传易感性关系的研究结果,所纳入文献均采用病例--对照研究的设计,以乳腺癌组与对照组人群基因型分布的OR值为效应指标,采用Rev Man4.2软件进行Meta-分析,并根据一致性检验的结果选用合适的模型(固定模型或随机模型)进行数据的合并。结果本次Meta-分析共纳入符合要求的文献7篇,其中英文5篇,中文2篇,累计病例数和对照数分别为1926例和3200例。CYP17c/c(纯合子模型)合并OR值为1.09(95%CL:0.91~1.30),Z值为0.93,相应的P值为0.35。CYP17c/t(杂合子模型)合并OR值为0.84(95%CL:0.64~1.12),Z值为1.19,相应的P值为0.23。其中5篇文献把所研究总体女性分成绝经前及绝经后人群,Meta-分析结果显示绝经前后均未发现存在相关性。结论尚无足够的证据证明CYP17基因多态性与中国女性乳腺癌易感性有确切关系。
Objective To comprehensively evaluate the association between CYP17 polymorphism and genetic predisposition of female breast cancer in China. Methods The Chinese and English databases of Wanfang, VIP, CNKI, Medline, springerlink and PUBMED were searched to obtain the relationship between CYP17 genetic polymorphism and genetic predisposition of breast cancer in Chinese women. All the included articles were studied by case-control study Design, the OR value of genotype distribution in breast cancer patients and control subjects as the effect indicators, using Meta-analysis Rev Man4.2 software, and the results of the consistency test using the appropriate model (fixed model or random model) Data consolidation. Results The Meta-analysis included 7 articles that met the requirements, including 5 articles in English and 2 articles in Chinese. The total number of cases and controls were 1926 cases and 3200 cases respectively. The combined OR of CYP17c / c (homozygous model) was 1.09 (95% CL: 0.91-1.30) with a Z value of 0.93 and a corresponding P value of 0.35. The combined odds ratio for CYP17c / t (heterozygote model) was 0.84 (95% CL: 0.64-1.12) with a Z value of 1.19 and a corresponding P value of 0.23. Among them, five articles divided the study population into pre-menopausal and post-menopausal groups. Meta-analysis showed no correlation existed before and after menopause. Conclusions There is not enough evidence to prove that the CYP17 gene polymorphism is closely related to the susceptibility of breast cancer in Chinese women.
