辣椒倍半萜植保素代谢的分子生态学研究 Ⅰ紫外线对辣椒叶片倍半萜环化酶及鲨烯合成酶的作用

为建立克隆辣椒倍半萜环化酶基因的cDNA文库,我们尝试了用紫外线(UV)处理离体辣椒叶片,考察UV对辣椒倍半萜环化酶活性的影响,结果表明:UV能诱导辣椒叶片表达信半萜环化酶活性;不同抗性水平的辣椒品种倍半萜环化酶活性对紫外线的反应有差异,紫外线处理后,抗病品种cm334在24h出现酶活性高峰,而感病品种subiche在36~42h出现酶活性高峰;在紫外线处理后0~24hcm334的酶活性高于subiche。UV对辣椒倍半萜环化酶活性的诱导作用与UV的作用方式、剂量及辣椒植株的生育期有关,紫外线交联处理45s诱导10叶期的倍半萜环化酶活性最高;5支15W紫外灯相距20cm照射15min低于1支相同的紫外灯处理;在相同的UV作用下,开花前辣椒叶片倍半萜环化酶活性显著高于开花以后的辣椒叶片。另一方面,紫外线处理是辣椒叶片中的鲨烯合成酶活性受到一定程度的拟制。 In order to establish a cDNA library of cloned sesquiterpene cyclase gene, we tried to treat the leaves of in vitro pepper by ultraviolet (UV) light and investigated the effect of UV on the sesquiterpene cyclase activity of pepper. The results showed that UV could induce pepper The expression of sesquiterpene cyclase was observed in leaves of different cultivars. The response of sesquiterpene cyclase activity to ultraviolet light of pepper cultivars with different resistance levels was different. After UV treatment, the peak of enzyme activity appeared in resistant cultivar cm334 at 24h, The peak of enzyme activity of subiche appeared at 36 ~ 42h, and the activity of enzyme at 0 ~ 24hcm334 after UV treatment was higher than that of subiche. The UV-induced sesquiterpene cyclase activity was related to the mode of action of UV, the dosage and the growth period of pepper plants. The highest sesquiterpene cyclase activity of 10-senescence stage was induced by UV-crosslinking 45s; UV lamp 20cm irradiation 15min less than 1 same UV lamp treatment; under the same UV effect, before flowering pepper sesquiterpene cyclase activity was significantly higher than after flowering pepper leaves. On the other hand, UV treatment is a function of squalene synthase activity in pepper leaves to some extent.
