目的:探讨pSilencer1.0-U6-siRNA-STAT3重组质粒抑制人卵巢癌SKOV3细胞裸鼠皮下移植瘤生长及诱导肿瘤细胞凋亡的作用。方法:应用人卵巢癌细胞株SKOV3对15只裸鼠构建卵巢癌移植瘤模型,随机分为生理盐水对照组、空质粒对照组、重组质粒组。联合电转染技术向裸鼠移植瘤内注射重组质粒,每隔5天重复注射1次,每4天测量各组裸鼠皮下移植瘤体积。采用TUNEL法检测肿瘤组织细胞的凋亡情况。结果:重组质粒瘤内注射对卵巢癌移植瘤生长有明显的抑制作用,与两个对照组比较重组质粒组裸鼠移植瘤生长速度明显减慢(P<0.01)。TUNEL染色显示重组质粒组癌组织有大量细胞凋亡。结论:pSilencer1.0-U6-siRNA-STAT3重组质粒能有效诱导人卵巢癌裸鼠皮下移植瘤组织中细胞凋亡,抑制肿瘤生长。
Objective: To investigate the effect of recombinant plasmid pSilencer1.0-U6-siRNA-STAT3 on the growth of subcutaneous xenografts and the induction of apoptosis of human ovarian cancer SKOV3 cells in nude mice. Methods: Ovarian cancer xenograft model was established in 15 nude mice with human ovarian cancer cell line SKOV3 and randomly divided into normal saline control group, empty plasmid control group and recombinant plasmid group. Recombinant plasmid was transplanted into nude mice by electroporating technique. The mice were injected once every 5 days and the volume of subcutaneous xenografts of nude mice in each group was measured every 4 days. TUNEL method was used to detect the apoptosis of tumor cells. Results: The recombinant plasmids intratumoral injection significantly inhibited the growth of ovarian cancer xenografts. Compared with the two control groups, the growth rate of transplanted tumor in nude mice significantly decreased (P <0.01). TUNEL staining showed that the recombinant plasmid had a large number of cancer cells apoptosis. Conclusion: The recombinant plasmid pSilencer1.0-U6-siRNA-STAT3 can effectively induce apoptosis and inhibit the growth of human ovarian cancer xenografts in nude mice.
