目的探讨多囊卵巢综合征(PCOS)患者卵巢颗粒细胞中雄激素相关microRNA的表达。方法将86例PCOS患者根据临床症状及血总睾酮浓度分为高雄激素PCOS组(47例)及非高雄激素PCOS组(39例),将同期50例因男方因素或者输卵管因素不孕者作为对照组。收集所有研究对象卵子,进行实时荧光定量PCR,分析数据并筛选差异表达的miRNAs,比较3组患者卵巢颗粒细胞中雄激素相关microRNA的表达差异。结果高雄激素PCOS组卵巢颗粒细胞中雄激素相关microRNA表达与非高雄激素PCOS组相比,多种microRNA的表达发生了变化。其中变化倍数在两倍以上的有17种microRNA(5种上调,12种下调)。与其他两组比较,hsa-miR-205、hsa-miR-30c两种上调micro RNA及hsa-miR-7、hsa-miR-181a、hsa-miR-25、hsamiR-943、hsa-miR-21、hsa-miR-668六种下调microRNA在高雄激素PCOS组卵巢颗粒细胞中变化显著(P<0.05)。结论PCOS患者卵巢颗粒细胞中hsa-miR-205、hsa-miR-7、hsa-miR-25等多种雄激素相关microRNA在其发病过程中发挥重要作用,为后续的临床治疗提供了靶点。
Objective To investigate the expression of androgen-related microRNA in ovarian granulosa cells of patients with polycystic ovary syndrome (PCOS). Methods Eighty-six patients with PCOS were divided into two groups: PCOS group (n = 47) and PCOS group (n = 39) according to the clinical symptoms and total testosterone concentration. Fifty patients with male PCOS or tubal infertility group. All the eggs were collected, real-time quantitative PCR was performed, the data were analyzed and the differentially expressed miRNAs were screened. The difference of expression of androgen-related microRNAs in ovarian granulosa cells was compared between the three groups. Results The expression of androgen-related microRNAs in ovarian granulosa cells of PCOS group was higher than that in non-hyperandrogenic PCOS group. Seventeen microRNAs (five up-regulated and twelve down-regulated) were found in which the fold change was more than twice. Compared with the other two groups, hsa-miR-205 and hsa-miR-30c upregulated the expression of microRNA and hsa-miR-7, hsa-miR- 181a, hsa- miR- 25, hsamiR- 943, hsa-miR- 21 , Hsa-miR-668 six kinds of down-regulated microRNA in ovarian granulosa cells of PCOS group had significant changes (P <0.05). Conclusions Various androgen-associated microRNAs such as hsa-miR-205, hsa-miR-7 and hsa-miR-25 in PCOS patients play an important role in the pathogenesis of ovarian granulosa cells and provide a target for subsequent clinical treatment.
