目的探讨HIF-Ⅰα及VEGF蛋白表达与食管鳞癌发生、发展及浸润、转移的关系。方法应用免疫组化SP法检测49例食管鳞癌组织及32例正常食管黏膜组织中HIF-Ⅰα及VEGF蛋白的表达。结果食管鳞癌组织中HIF-Ⅰα蛋白表达与癌组织的浸润深度及淋巴结转移密切相关(P<0.05),而与分化程度无关(P>0.05);VEGF蛋白表达与癌的组织学分级、浸润深度无关(P>0.05)而只与淋巴结转移有关(P<0.05);在食管鳞癌组织中HIF-Ⅰα与VEGF蛋白的阳性表达率分别为51.0%、67.3%,均明显高于相应正常黏膜组织中的阳性表达率0.0%、0.0%,(P<0.05)。HIF-Ⅰα与VEGF蛋白的表达呈正相关关系。结论HIF-Ⅰα和VEGF在食管鳞癌的浸润、转移及黏膜上皮癌变中起重要作用,联合检测HIF-Ⅰα及VEGF可作为判定食管鳞癌侵袭转移能力的客观指标,对食管鳞癌的预后判断具有重要意义。
Objective To investigate the relationship between the expression of HIF-1a and VEGF and the occurrence, development, invasion and metastasis of esophageal squamous cell carcinoma. Methods Immunohistochemical SP method was used to detect the expression of HIF-Ⅰα and VEGF in 49 cases of esophageal squamous cell carcinoma and 32 cases of normal esophageal mucosa. Results The expression of HIF-Ⅰα in esophageal squamous cell carcinoma was closely related to the depth of invasion and lymph node metastasis (P <0.05), but not to the degree of differentiation (P> 0.05). The expression of VEGF protein was correlated with the histological grade, (P> 0.05), but only related to lymph node metastasis (P <0.05). The positive rates of HIF-la and VEGF protein in esophageal squamous cell carcinoma tissues were 51.0% and 67.3%, respectively, which were significantly higher than those in corresponding normal mucosa The positive expression rate in tissues was 0.0%, 0.0% (P <0.05). There was a positive correlation between the expression of HIF-la and VEGF protein. Conclusions HIF-Ⅰα and VEGF play an important role in the invasion and metastasis of esophageal squamous cell carcinoma and mucosal epithelial carcinogenesis. Combined detection of HIF-Ⅰα and VEGF can be used as an objective index to determine the invasion and metastasis of esophageal squamous cell carcinoma.
