目的:通过观察田麻益智颗粒含药血清对GLu损伤的胎鼠海马神经元的保护作用,探讨田麻益智颗粒对脑损伤神经元的保护机制。方法:取孕7 d Wistar大鼠的胎鼠海马,进行体外培养,并对培养的神经干细胞进行神经元鉴定。部分细胞开始贴壁,贴壁细胞呈圆形结果培养24 h后,大部分细胞己经贴壁生长:分化3 d后神经元胞体增大,突起延长并出现分枝,形成稀疏的网络,7d胞体大,突起长,分枝连接成网,相互联系增多细胞成熟。MTT结果表明,田麻益智颗粒含药血清保护组与Glu损伤模型组相比细胞活性明显升高(P<0.05)。平均值,田麻益智颗粒中剂量组对Glu损伤的神经元细胞保护作用与尼莫地平组的保护作用接近,优于正常组及模型组。说明田麻益智颗粒含药血清对Glu损伤海马神经元有保护作用。免疫荧光结果半定量分析结果标记CAMKII的神经元细胞中,统计分析结果表明正常组平均灰度值为148.6±9.88、保护组灰度值151.0±12.37低于Glu损伤组171.5±6.87(P<0.05),说明保护组中CAMKII的含量与正常组接近,并且较Glu损伤模型组少,中剂量组田麻益智颗粒对谷氨酸损伤神的海马神经元的有较好的保护作用。
OBJECTIVE: To observe the protective effect of Tianma Yizhi granule-containing serum on GLu injured fetal rat hippocampal neurons and to explore the protective mechanism of TIANMA Yizhi granule on neurons injured by GLu. Methods: Fetal rat hippocampus of 7-day-old Wistar rats were cultured in vitro and the neurons were identified. Some cells started adherent and adherent cells were round. After cultured for 24 h, most of the cells had adherent growth. After 3 days of differentiation, the cell bodies of neurons increased, and the protrusions extended and branched to form a sparse network. After 7 days Large cell body, protuberances, branches connected into a network, increased mutual maturity of cells. The results of MTT showed that the cell activity of Tianma Yizhi granule drug-containing serum protection group was significantly higher than that of Glu injury model group (P <0.05). Mean, Tianma Yizhi granules medium dose group on Glu injury neuronal protective effect and nimodipine group close, better than normal group and model group. Description Tianma Yizhi granule containing serum on Glu injury hippocampal neurons have a protective effect. Results of semi-quantitative analysis of immunofluorescence showed that CAMKII-labeled neuron cells showed that the average gray value of normal group was 148.6 ± 9.88 and that of protective group was 151.0 ± 12.37, lower than that of Glu group (171.5 ± 6.87, P <0.05) ), Indicating that the content of CAMKII in the protection group is close to that of the normal group and less than that of the Glu injury model group. The middle-dose Tianma Yizhi Granule has a good protective effect on the hippocampal neurons damaged by glutamic acid.
