目的研究药用植物香鳞毛蕨核糖体18SrRNA基因的序列特征。方法以香鳞毛蕨总RNA为模板,根据蕨类植物18S rRNA保守区域设计引物,对香鳞毛蕨18SrRNA基因进行基因克隆、测序、并进行序列比对和同源性分析。结果克隆获得香鳞毛蕨18S rRNA 1010bp部分片段(Gen Bank登录号GU385474),进行Blast对比发现其与大羽鳞毛蕨的同源性达到99%。结论 DNA测序技术可为香鳞毛蕨的鉴定提供准确而有效的分子鉴定方法,也可为其药用成分相关基因的研究提供基础性资料。
Objective To study the sequence characteristics of the ribosomal 18S rRNA gene of Phyllostachys pubescens from medicinal plants. Methods The total RNA was used as a template, and the 18S rRNA gene was cloned and sequenced according to the conservative region of ferns 18S rRNA. Sequence alignment and homology analysis were performed. Results A partial fragment of 18S rRNA 1010bp (Gen Bank Accession No. GU385474) was obtained by cloning. The Blast comparison showed that the homology of the gene to the Pseudora fimbriatus reached 99%. Conclusion DNA sequencing technology can provide an accurate and effective molecular identification method for the identification of Fraxinus mandshurica, and also provide basic information for the study of related genes of medicinal components.
