目的 探讨沙立度胺(Thd;商品名:反应停)对人卵巢癌裸鼠皮下移植瘤生长及血管生成的影响。方法 建立 15只人卵巢癌裸鼠皮下移植瘤模型,随机分成 3组: (1)Thd组:腹腔内注射Thd(200mg·kg-1·d-1,浓度为 62 5mg/L); (2 )Thd+环磷酰胺 (CTX)组:按上述Thd剂量,加入浓度为 100mg/L的CTX(100mg·kg-1·d-1 ),腹腔内注射; (3)生理盐水组:腹腔内注射 0 2ml生理盐水。观察各组裸鼠皮下移植瘤生长情况、裸鼠体重的变化,并采用RT PCR、酶联免疫吸附法、及Picture二步免疫组化法检测各组瘤组织中血管内皮生长因子 (VEGF)mRNA、外周血中VEGF含量及瘤组织中微血管密度(MVD)。结果 ( 1 )Thd组、Thd+CTX组皮下移植瘤体积明显小于生理盐水组,差异有统计学意义(P<0.05)。(2)Thd组瘤组织中VEGFmRNA为 55±9,血中VEGF含量为(29±10)pg/ml;Thd+CTX组瘤组织中VEGFmRNA为 26±7,血中VEGF含量为(12 ±6)pg/ml;生理盐水组瘤组织中VEGFmRNA为 79±7,血中VEGF含量为(71±16)pg/ml。Thd组、Thd+CTX组瘤组织中VEGFmRNA和血中VEGF含量分别与生理盐水组比较,差异均有统计学意义 (P<0.01)。(3)Thd组MVD计数为 (12.6±3.3)条,Thd+CTX组为 (10.6±1.9)条,均明显低于生理盐水组的(19 3±2 8)条(P<0 01)。结论 Thd有抗卵巢癌皮下移植瘤血
Objective To investigate the effect of thalidomide (Thd; trade name: reaction stop) on the growth and angiogenesis of subcutaneous xenografts of human ovarian cancer in nude mice. Methods Fifteen human ovarian cancer xenografts were established and divided into three groups at random: (1) Thd group: intraperitoneal injection of Thd (200 mg · kg-1 · d-1, concentration of 62 5 mg / L) ) Thd + cyclophosphamide (CTX) group: CTX (100mg · kg-1 · d-1) with a concentration of 100mg / L was injected intraperitoneally at the Thd dose; (3) saline group: intraperitoneal injection of 0 2ml saline. The growth of subcutaneous xenografts in nude mice and body weight of nude mice in each group were observed. RT-PCR, enzyme-linked immunosorbent assay (ELISA) and two-step immunohistochemistry were used to detect the expression of vascular endothelial growth factor (VEGF) , VEGF in peripheral blood and microvessel density (MVD) in tumor tissues. Results (1) The volume of subcutaneous tumor in Thd group and Thd + CTX group was significantly smaller than that in saline group (P <0.05). (2) The VEGF mRNA in Thd group was 55 ± 9 and that in blood was (29 ± 10) pg / ml. The VEGF mRNA in Thd + CTX group was 26 ± 7 and the level of VEGF in blood was (12 ± 6) ) pg / ml; the VEGF mRNA in the normal saline group was 79 ± 7, and the blood VEGF level was (71 ± 16) pg / ml. The levels of VEGF mRNA and VEGF in Thd group and Thd + CTX group were significantly different from those in normal saline group (P <0.01). (3) The MVD counts in Thd group were (12.6 ± 3.3) and in Thd + CTX group (10.6 ± 1.9), respectively, which were significantly lower than those in normal saline group (19 3 ± 28) (P <0.01).
