本实验用酶组织化学法探讨新鲜芦荟汁对四氯化碳诱发损伤的培养肝细胞的保护作用。将Wistar系大白鼠乳鼠的肝细胞原代单层培养4~5天,取生长良好的培养瓶分为正常组、损伤组、实验组。在各培养瓶中取肝细胞生长盖玻片,显示琥珀酸脱氢酶(SDH)、酸性磷酸酶(ACP)、糖原(PAS反应)等。结果损伤组肝细胞比正常组肝细胞琥珀酸脱氢酶流行性明显降低,酸性磷酸酶活性明显增强。糖原显示反应结果,糖原颗粒明显减少.但实验组琥珀酸脱氢酶和酸性磷酸酶活性及糖原显示反应结果均与正常组相似。实验结果提示新鲜芦荟汁对损伤肝细胞具有良好的保护作用。
In this study, enzymatic histochemistry was used to investigate the protective effect of fresh aloe vera juice on cultured hepatocytes induced by carbon tetrachloride. Primary cultured hepatocytes of Wistar rats were cultured for 4 to 5 days. The well-growing culture flasks were divided into normal group, injury group and experimental group. Hepatocyte growth coverslips were taken from each flask and showed succinate dehydrogenase (SDH), acid phosphatase (ACP), glycogen (PAS reaction) and the like. Results Compared with normal group, the hepatocytes in the injury group had a significantly lower prevalence of succinate dehydrogenase and a significantly higher acid phosphatase activity. Glycogen showed the reaction results, glycogen particles significantly reduced. But the experimental group succinate dehydrogenase and acid phosphatase activity and glycogen showed that the reaction results are similar to the normal group. Experimental results suggest that fresh aloe vera juice has a good protective effect on injured liver cells.
