目的:观察小剂量重复照射下不同时相大鼠血清内皮素(ET-1)和肿瘤坏死因子-α(TNF-α)的改变并探讨益气活血中药对放射性肺损伤的防治作用。方法:将96只W istar雌性大鼠随机分为照射加中药组(A组)30只、照射加氨溴索组(B组)30只、单纯照射组(C组)30只、正常对照组(D组)6只。用直线加速器对前三组大鼠右肺进行分次照射(5 Gy/次,1次/周,累积剂量最高为30 Gy),于照射开始后第4、6、8、12、26周5个时间点分别处死A、B、C组各6只大鼠,取血清标本进行ELISA检测,观察各组大鼠不同时相血清ET-1和TNF-α的动态变化。结果:3组血清ET-1水平于照射后4周进行性升高,8周达高峰,12周开始下降;3组血清TNF-α水平4周进行性升高,12周达高峰,26周开始下降。A组各时相大鼠血清ET-1、TNF-α表达水平均低于C组(P<0.05),且各时相值与B组近似,差异无统计学意义(P>0.05)。结论:血清ET-1、TNF-α水平的变化能反映早期放射性肺损伤的严重程度,益气活血中药能够降低血清ET-1、TNF-α的水平,可能是其防治放射性肺损伤的作用机制。
OBJECTIVE: To observe the changes of serum endothelin-1 (ET-1) and tumor necrosis factor-α (TNF-α) in rats with different doses of repeated irradiation and to investigate the preventive and therapeutic effects of Yiqihuoxue herbs on radiation-induced lung injury. METHODS: Ninety-six Wistar female rats were randomly divided into three groups: irradiation plus Chinese medicine group (group A), irradiation plus ambroxol group (group B), and simple irradiation group (group C), normal control group. (Group D) 6 individuals. The right lungs of the first three groups were irradiated by a linear accelerator (5 Gy/time, 1 time per week, cumulative dose up to 30 Gy), at the 4th, 6th, 8th, 12th, and 26th weeks after irradiation began. Six rats in groups A, B, and C were sacrificed at each time point. Serum samples were taken for ELISA and the dynamic changes of serum ET-1 and TNF-α at different phases were observed in each group. Results: The levels of serum ET-1 in three groups increased progressively 4 weeks after irradiation, peaked at 8 weeks and began to decrease at 12 weeks. Serum TNF-α levels in 3 groups increased progressively at 4 weeks, peaked at 12 weeks, and reached 26 weeks. begin descending. The levels of serum ET-1 and TNF-α in each phase of group A were significantly lower than those in group C (P<0.05), and the values of each phase were similar to those of group B. There was no significant difference (P>0.05). Conclusion: The changes of serum ET-1 and TNF-α levels can reflect the severity of early radiation-induced lung injury. Yiqihuoxue herbs can reduce the levels of serum ET-1 and TNF-α, which may be the mechanism of prevention and treatment of radiation-induced lung injury. .
