目的观察久泻灵冲剂对溃疡性结肠炎大鼠模型的影响,为临床用药的有效性提供实验依据。方法采用异种异体结肠黏膜组织致敏法和乙酸局部灌肠相结合的方法制造动物模型。将Wistar大鼠随机分为空白对照组、模型对照组、阳性对照组及久泻灵冲剂大、中、小剂量组。连续给药14 d,末次给药后24 h处死大鼠,制备结肠组织匀浆,生化法检测结肠组织中GSH-Px、SOD、MDA的含量,结肠组织HE染色后进行病理学组织评分。结果久泻灵可明显降低结肠组织中MDA的活性水平,升高GSH-Px、SOD的活性,降低病理组织学评分。结论久泻灵冲剂对溃疡性结肠炎模型大鼠有显著的干预作用,其机制可能与抗氧自由基损伤有关。
Objective To observe the effects of Jiuxianling granules on ulcerative colitis in rats and provide experimental evidence for its clinical efficacy. Methods Animal model was made by sensitization of allogeneic colonic mucosa tissue and local acetic acid enema. Wistar rats were randomly divided into blank control group, model control group, positive control group and Jisanling Granules in large, medium and small dose groups. The rats were sacrificed 24 h after the last administration, and the colon tissue homogenate was prepared. The contents of GSH-Px, SOD and MDA in the colon tissues were determined by biochemical method. The pathological tissues were scored after HE staining. Results Jiu Xie Ling can significantly reduce the activity of MDA in colon tissue, increase the activity of GSH-Px and SOD, and reduce the histopathological score. Conclusion Jiuxiaoling granules has a significant intervention on ulcerative colitis model rats, and its mechanism may be related to the antioxidant free radical damage.
