目的:探讨微粒子酶免疫分析法测他克莫司血药浓度时样品的保存时间、保存条件,以及不同抗凝剂对测定结果的影响。方法:取肝肾移植术后患者静脉全血标本,分别测定室温和冰箱保存3d、7d的血药浓度,以考察温度对测定结果的影响;测定采用不同抗凝剂的标本血药浓度以考察抗凝剂对测定结果的影响。结果:室温或冰箱冷藏保存后标本测定结果与保存前的测定值无统计学差异;EDTA-K2与肝素钠采集的标本2组间无统计学差异。结论:血样在室温下至少可保存1周;采血管可以选用EDTA-K2或肝素钠抗凝,二者差异无显著性。
OBJECTIVE: To investigate the preservation time, storage conditions, and the effect of different anticoagulants on the determination results when measuring the concentration of tacrolimus in serum by particle enzyme immunoassay. Methods: The venous blood samples of patients after liver-kidney transplantation were collected, and the plasma concentrations at room temperature and in refrigerators at 3d and 7d respectively were measured to investigate the influence of temperature on the determination results. Results: There was no significant difference between the results of pre-preservation and frozen-thawed samples at room temperature or in refrigerator. There was no significant difference between EDTA-K2 and heparinized samples. Conclusion: Blood samples can be stored for at least 1 week at room temperature. EDTA-K2 or heparin sodium anticoagulation can be used in blood collection tubes. There is no significant difference between the two methods.
