目的建立HPLC同时测定连翘中连翘苷、芦丁和槲皮素的含量。方法采用ZORBAX SB C18(4.6 nm×250 mm,5μm)色谱柱;以甲醇-0.2%磷酸水溶液为流动相,梯度洗脱(0~40 min,40%~50%甲醇);连翘苷检测波长为280 nm,芦丁和槲皮素检测波长为370 nm;流速为1.0 mL.min-1;柱温为25℃。结果 3个成分均达到良好分离,连翘苷、芦丁和槲皮素平均加样回收率分别为98.2%,99.7%,100.9%。结论本检测方法简便,准确性和重现性良好,可以作为同时测定连翘中连翘苷、芦丁和槲皮素含量的方法。
OBJECTIVE To establish HPLC simultaneous determination of forsythin, rutin and quercetin in Forsythia suspensa. Methods The mobile phase of ZORBAX SB C18 (4.6 nm × 250 mm, 5 μm) was used as mobile phase. The mobile phase consisted of methanol-0.2% phosphoric acid solution with gradient elution (0-40 min, 40% -50% methanol) At 280 nm, the detection wavelength of rutin and quercetin was 370 nm, the flow rate was 1.0 mL.min-1 and the column temperature was 25 ℃. Results All the three components were well separated. The average recoveries of forsythin, rutin and quercetin were 98.2%, 99.7% and 100.9%, respectively. Conclusion The method is simple, accurate and reproducible. It can be used as a method for simultaneous determination of forsythin, rutin and quercetin in Forsythia suspense.
