目的揭示香豆素类成分在川白芷药用部位根中的分布及相对含量与根直径的关系;同时,为中药组织化学研究方法提供一定的参考。方法以川白芷新鲜根为研究对象,运用冰冻切片以及高内涵细胞分析技术。结果川白芷根冰冻切片在高内涵细胞分析系统下,分泌道清晰,香豆素的自发荧光激发效果好,能科学记录各切片的荧光平均强度,反映出川白芷根中香豆素类成分的相对含量;随着白芷根横切片的直径增大,其分泌道中香豆素自发荧光强度并未呈现线性式增强。结论高内涵细胞分析系统能用于白芷根中香豆素类成分的定位及相对定量研究。川白芷新鲜根中香豆素的相对含量与直径并不呈现正相关关系。
Objective To reveal the distribution and relative content of coumarins in the roots of Chuanzhihizhizhi (Radix Angelicae Sinensis) in relation to the root diameter, and to provide some references for the histochemical methods of traditional Chinese medicine. Methods Chuanbaizhi fresh roots as the research object, the use of frozen sections and high content of cell analysis technology. Results Frozen sections of Radix Angelicae Sinensis had clear secretions under high-content cell analysis system and good autofluorescence excitation of coumarin. It could scientifically record the average fluorescence intensity of each section and reflect the relative content of coumarins in Radix Angelicae Sinensis Content; with Angelica root cross-section diameter increases, the secretion of coumarin spontaneous fluorescence intensity did not show a linear increase. Conclusion The high content of cell analysis system can be used for the localization and relative quantification of coumarins in Angelica root. The relative content of coumarins in fresh roots of Chuan Baizhi did not show a positive correlation with the diameter.
