目的建立同时测定复方红花喷雾剂中羟基红花黄色素A(hydroxysafflor yellow A,HSYA)与绿原酸含量的方法。方法采用双波长HPLC,色谱柱:Shim-pack VP-ODS(250 mm×4.6 mm,5μm),流动相:乙腈-0.4%磷酸溶液(12∶88),流速为1.0 m L·min?1,检测波长:403 nm(HSYA)和327 nm(绿原酸),柱温为30℃。结果 HSYA、绿原酸的浓度分别在0.612~12.24,0.632~12.64μg·m L?1内与各自峰面积呈良好的线性关系(r均为0.999 9);平均加样回收率分别为98.34%和99.82%,RSD分别为1.16%和1.64%(n=9)。结论该方法操作简便、快速,结果可靠,具有良好的重复性,可用于复方红花喷雾剂中HSYA与绿原酸的含量测定。
Objective To establish a method for the simultaneous determination of hydroxysafflor yellow A (HSYA) and chlorogenic acid in Fufang Honghua Spray. Methods The two-wavelength HPLC was used. The chromatographic column was Shim-pack VP-ODS (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of acetonitrile and 0.4% phosphoric acid solution Detection wavelength: 403 nm (HSYA) and 327 nm (chlorogenic acid), the column temperature was 30 ℃. Results The concentrations of HSYA and chlorogenic acid were linear with their peak areas in the range of 0.612 ~ 12.24 and 0.632 ~ 12.64 μg · m L -1, respectively (r = 0.999 9). The average recoveries of HSYA and chlorogenic acid were 98.34% And 99.82% with RSDs of 1.16% and 1.64%, respectively (n = 9). Conclusion The method is simple, rapid, reliable and has good reproducibility. It can be used for the determination of HSYA and chlorogenic acid in compound safflower spray.
