目的观察转化生长因子6(TGF-β)对培养的人角膜基质细胞分泌MMP-2和MMP-9的调节作用,探讨TGF-β治疗非感染性角膜溃疡的潜在可能性。方法采用酶谱法检测人角膜基质细胞MMP-2和MMP-9的分泌,并观察 TGT-β对 MMP-2和 MMP-9分泌的调节作用。结果培养的人角膜基质细胞分泌 MMP-2和 MMP-9,TGF-β抑制MMP-9的活性,且随TGF-β质量浓度增加而增强,随TGF-β作用时间延长而增强;对MMP-2的活性,TGF-β有较弱的促进作用。结论TGF-β作为一种MMP-9的抑制剂,在促进非感染性角膜溃疡愈合方面很有潜力。
Objective To investigate the regulatory effect of transforming growth factor-6 (TGF-β) on secretion of MMP-2 and MMP-9 by cultured human corneal stromal cells and to explore the potential of TGF-β in the treatment of non-infected corneal ulcer. Methods The secretion of MMP-2 and MMP-9 in human corneal stroma cells was detected by zymography, and the regulation of the secretion of MMP-2 and MMP-9 by TGT-β was observed. Results The cultured human corneal stromal cells secreted MMP-2 and MMP-9, the activity of TGF-β inhibited the activity of MMP-9 and increased with the increase of TGF-β concentration, 2 activity, TGF-β has a weaker role in promoting. Conclusion TGF-β, as a MMP-9 inhibitor, has great potential in promoting the healing of non-infectious corneal ulcer.
