用免疫组织化学方法检测40例原发性肝癌及其癌周肝组织中突变型p53基因蛋白(mp53)和增殖细胞核抗原(PCNA)的表达.结果表明:mp53和PCNA的表达均定位于细胞核内,表达分布是弥漫、局灶、散在3种表现.其阳性表达率:在原发性肝癌(PHC)中,mp53蛋白为45.0%(18/40),PCNA为72.5%(29/40);癌周肝组织中,mp53蛋白为2.5%(1/40),PCNA为40.0%(16/40);癌周肝硬变中mp53蛋白为11.1%(1/9),PCNA为33.3%(3/9).结果提示:mp53蛋白主要是在肿瘤细胞中表达的标志物,并能反映癌周肝细胞中存在少量癌细胞的转移;PCNA可能是各种不同细胞异常增殖处于不同细胞周期阶段很有价值的标记物.mp53基因蛋白和PCNA对原发性肝癌的发生和发展可能有重要的影响.mp53蛋白可能与正常肝细胞的恶性转化有较密切的关系,涉及一些包括PCNA以及有关细胞增殖和分化的基因调控在内的其它多种因素有关.
Immunohistochemistry was used to detect the expression of mutant p53 gene protein (mp53) and proliferating cell nuclear antigen (PCNA) in 40 cases of primary liver cancer and its surrounding liver tissue. The results showed that the expression of both mp53 and PCNA was localized in the nucleus. The expression distribution was diffuse, focal and scattered in three manifestations. The positive expression rate was: in primary liver cancer (PHC), mp53 protein was 45.0% (18/40), and PCNA was 72.5% (29/40); In the liver tissue around the cancer, mp53 protein was 2.5% (1/40), PCNA was 40.0% (16/40), peritoneal cirrhosis had mp53 protein of 11.1% (1/9), and PCNA was 33.3% (3 /9). The results suggest that: mp53 protein is mainly expressed in tumor cells and can reflect the presence of a small amount of cancer cells in the hepatic cancer cells; PCNA may be abnormal proliferation of different cells in different cell cycle stages. The valuable marker mp53 gene protein and PCNA may have important influence on the occurrence and development of primary liver cancer. The mp53 protein may have a close relationship with the malignant transformation of normal hepatocytes, involving some including PCNA and related cell proliferation. It is related to many other factors including differentiation of gene regulation.
