目的研究活血祛瘀中药对SiO2所致矽肺模型肺组织中转化生长因子-β1(TGF-β1)、肿瘤坏死因子-α(TNF-α)及白介素-6(IL-6)表达的影响。方法将SD大鼠随机分为6组,分别为SiO2组、强的松+SiO2组、中药+SiO2组、强的松组、中药组、生理盐水组。末次给药后4 h,苯二胺盐酸盐法测定血清铜蓝蛋白(CP)含量;取肺组织,HE染色法行组织病理观察,免疫组化法测定肺组织肺组织胶原蛋白、TGF-β、TNF-α及IL-6的表达情况。结果治疗组与NS灌胃组病理检查未见明显差别;CP检测结果,SiO2+中药组较SiO2组、SiO2+西药组略有下降,但只与SiO2组比较差异有统计学意义;免疫组化法测定中药与对照组、西药治疗组肺组织胶原蛋白、TGF-β、TNF-α及IL-6的表达差异无统计学意义。结论在矽肺较严重的情况下,该抗矽肺方剂可能不能对矽肺模型起到干预的作用。
Objective To study the effects of traditional Chinese medicine for promoting blood circulation and removing blood stasis on the expression of transforming growth factor-β1, tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in the lungs of silicosis model induced by SiO2. Methods SD rats were randomly divided into 6 groups: SiO2 group, prednisone + SiO2 group, TCM + SiO2 group, prednisone group, TCM group and NS group. 4 h after the last administration, the content of ceruloplasmin (CP) was measured by phenylenediamine hydrochloride method. The lung tissue and HE staining were used to observe the histopathology. The lung tissue collagen, TGF- β, TNF-α and IL-6 expression. Results There was no significant difference in the pathological examination between the treatment group and the NS-fed group. The results of the CP test showed that there was a slight difference between the SiO2 + TCM group and the SiO2 group and the SiO2 + There was no significant difference in the expression of collagen, TGF-β, TNF-α and IL-6 between the Chinese medicine group and the control group and western medicine group. Conclusion In the more severe case of silicosis, the anti-silicosis prescription may not play an intervention role in the silicosis model.
