目的观察复方中药糖耐康对高糖环境下大鼠系膜细胞外基质的纤维连接蛋白(FN)、Ⅳ型胶原蛋白(IV-C)及转化生长因子TGF-β1含量的影响。方法大鼠肾小球系膜细胞按1×105/孔接种24孔板,实验分为6组:正常葡萄糖组、高糖对照组、中药糖耐康高、中、低浓度干预组、西药吡格列酮组。每组设平行孔4孔,培养24 h后检测指标。采用双抗夹心酶联免疫吸附(ELISA)法检测FN、Ⅳ-C及TGF-β1含量。结果与空白对照组比较,高糖模型组MCs的上清中的FN、Ⅳ-C、TGF-β1含量均显著升高,差异有统计学意义(P均<0.05);与高糖组比较,治疗组FN、Ⅳ-C、TGF-β1含量均显著降低,差异有统计学意义(P<0.01);与吡格列酮组比较,糖耐康中、低剂量组FN、IV-C、TGF-β1含量差异有统计学意义(P<0.01),而糖耐康高剂量组各指标差异无统计学意义(P>0.05)。结论糖耐康对糖尿病肾病肾脏的保护作用之一,是通过抑制系膜细胞外基质中FN、Ⅳ-C、TGF-β1的分泌、产生,从而抑制了系膜细胞的增值,起到保护糖尿病患者肾脏的作用。
Objective To observe the effects of Tangkang Kang, a compound Chinese herbal medicine, on the content of FN, IV-C and TGF-β1 in rat mesangial extracellular matrix (ECM) under high glucose condition. Methods Rat mesangial cells were seeded into 24-well plates at a density of 1 × 105 / well. The experiment was divided into 6 groups: normal glucose group, high glucose control group, traditional Chinese medicine glucose tolerance group, medium and low concentration intervention group, western medicine pioglitazone group. Each set of parallel holes 4 holes, 24 h after the test indicators. The contents of FN, Ⅳ-C and TGF-β1 were detected by enzyme-linked immunosorbent assay (ELISA). Results Compared with the blank control group, the contents of FN, Ⅳ-C and TGF-β1 in supernatants of MCs in high glucose group were significantly increased (all P <0.05). Compared with the high glucose group, Compared with pioglitazone group, the content of FN, IV-C and TGF-β1 in the middle and low dose of Tangjikang group were significantly decreased (P <0.01) The differences were statistically significant (P <0.01), while the high-dose sugar Kangkang no significant difference between the various indicators (P> 0.05). CONCLUSION: The protective effect of Tangnaikang on the kidney of diabetic nephropathy is through inhibiting the secretion of FN, Ⅳ-C and TGF-β1 in the extracellular matrix of mesangial cells, thereby inhibiting the proliferation of mesangial cells and protecting the diabetes The role of the patient’s kidneys.
