目的:考察老年哮喘患者口服氨茶碱常规监测群体药动学特征,为临床调整个体化给药方案提供依据。方法:回顾性收集我院117例老年哮喘患者的237份氨茶碱血清浓度样本,建立氨茶碱群体分析数据库,用非线性混合效应模型法(NONMEM)进行模型优化,分别考察性别、年龄、身高等因素对药动学参数的影响,建立最终模型方程。结果:氨茶碱数据用群体药动学模型进行描述,年龄和是否为多次给药对药动学参数有显著性影响。结论:该方法精准可靠,可用于临床调整给药方案。
OBJECTIVE: To investigate the pharmacokinetics of aminophylline in elderly patients with asthma and to provide evidence for clinical adjustment of individualized drug delivery programs. Methods: A total of 237 aminophylline serum samples from 117 elderly patients with asthma were retrospectively collected. The aminophylline database was established and the model was optimized by nonlinear mixed effect model (NONMEM). The effects of gender, age, Height and other factors on the pharmacokinetic parameters, the establishment of the final model equation. Results: Aminophylline data were described using a population pharmacokinetic model. Age and multiple drug administration had significant effects on pharmacokinetic parameters. Conclusion: The method is accurate and reliable, and can be used to adjust the dosing regimen.
