目的:探讨黄竹清脑颗粒对痰热腑实证脑出血模型大鼠急性期脑组织中半胱氨酸蛋白酶(Caspase-3)表达的影响。方法:采用SD大鼠自体粪便灌胃配合胶原酶Ⅶ-肝素混合液立体定位注射的方法复制痰热腑实证脑出血大鼠模型,免疫组化法测定大鼠脑组织中Caspase-3表达的变化。结果:①黄竹清脑颗粒可明显减轻痰热腑实证脑出血大鼠喉中痰鸣、鼻分泌物多,粪便干结,烦躁等症状,并可改善大鼠脑出血后的神经体征;②黄竹清脑颗粒大、中剂量组明显优于清开灵颗粒组(P<0.05)。结论:黄竹清脑颗粒能降低大鼠脑出血后血肿周围神经元活化caspase-3的表达,阻止神经元的凋亡,可能是其保护脑神经细胞的作用机理之一。
Objective: To investigate the effect of Huangzhu Qingnao granule on the expression of caspase-3 in brain tissue of rats with phlegm-heat-induced positive intracerebral hemorrhage model. Methods: SD rats were injected intrarectally with collagenase VII-heparin mixture by autologous fecal administration, and then the rats were sacrificed. The changes of Caspase-3 expression in rat brain were detected by immunohistochemistry . Results: Huangzhu Qingnao granule can significantly reduce the symptoms of phlegm, nasal secretions, dry stool, irritability and other symptoms in the larynx in rats with phlegm-heat syndrome, and can improve the neurological signs after intracerebral hemorrhage in rats. The large and middle dose groups were significantly better than Qingkailing granules group (P <0.05). Conclusion: Huangzhuqingnao Granule can reduce the expression of activated caspase-3 in the perihematoma after ICH and prevent the apoptosis of neurons, which may be one of the mechanisms of its protection.
