目前鉴定线虫种类的依据之一是测量线虫各部位的长宽尺度。测量线虫时,不管采用德曼氏(De Man)公式或采用柯柏氏(Cobb)公式,都得按需要测量线虫各部位的长宽尺度。目前国内外测量线虫时都是采用测微尺测量,而线虫死后多呈各种弯曲状,有的甚至卷成时钟的发条状。用直的测微尺在显微镜下测量弯的线虫,难度是很大的,不但速度慢,而且很不准确。测量线虫时不可能用测微尺量一次便达到目的,需要逐次测量逐次累加。每测量完一次又不可能在线虫身上划一个记号,在移动线虫以便下次接着测量的过程中,很可能将已量过的地方重量,或将没有量过的地方漏量,造成测量上的差错,导致鉴定上的错误。本文要谈及的是测量线虫的新技术。
