本研究从细胞周期调控这一角度来探讨胆固醇缺乏对T淋巴细胞增殖影响的分子机制。结果显示:经去脂血清培养基培养的Jurkat细胞加lovastatin 处理3d 后,3H-TdR掺入率明显下降,免疫细胞化学染色及免疫印迹法分析表明处理后的细胞cyclin D1、CDK4及pRb 蛋白的表达明显降低,加入低密度脂蛋白(LDL)可以部分逆转上述变化。结果提示,无论是内源性还是外源性胆固醇的缺乏都能抑制Jurkat细胞的增殖,推测这种变化可能与cyclin D1、CDK4及pRb 蛋白表达量的下降而导致相应的cyclin D1/CDK4复合活性下降有关
This study explored the molecular mechanism of the effects of cholesterol deficiency on T lymphocyte proliferation from the perspective of cell cycle regulation. The results showed that 3H-TdR incorporation decreased significantly in Jurkat cells treated with devastated serum for 3 days, and the incorporation rate of 3H-TdR decreased significantly. Immunocytochemistry and Western blot analysis showed that the cyclin D1, CDK4 and pRb proteins The expression was significantly reduced, adding low-density lipoprotein (LDL) can partially reverse the above changes.
