目的:观察益气解毒活血方对NZB/W F1狼疮鼠肾炎肾纤维化的干预作用,并探讨其可能的作用机制。方法:随机将5月龄狼疮鼠分为空白对照组(空白组)、益气解毒活血小复方治疗组(中药组)、骁悉治疗组(西药组)、益气解毒活血小复方+骁悉治疗组(中西组)。持续干预治疗8周,并分别于实验第0、2、4、6周收集狼疮鼠24 h尿液进行蛋白定量检测,干预结束后观察不同组别狼疮鼠肾脏病理、肾脏羟脯氨酸(Hyp)含量及肾组织转化生长因子-β_1(TGF-β_1)、结缔组织生长因子(CTGF)mRNA的表达情况。结果:不同治疗组均对NZB/W F1狼疮鼠24 h尿蛋白定量、肾脏Hyp含量、肾脏病理、肾组织TGF-β_1与CTGF mRNA表达有改善作用,但各治疗组之间比较无统计学差异;而肾脏病理显示益气解毒活血小复方+骁悉治疗组肾脏病变程度最轻。结论:益气解毒活血方具备降低NZB/W F1狼疮鼠蛋白尿,改善肾纤维化作用,其机制可能与下调致纤维化因子TGF-β_1、CTGF mRNA表达相关。
Objective: To observe the intervention of Yiqi Jiedu Huoxue decoction on nephritis renal fibrosis in NZB / W F1 wolf mice and to explore its possible mechanism. Methods: 5-month-old lupus mice were randomly divided into control group (blank group), Yiqi Jiedu Huoxue Compound Prescription group (Chinese medicine group), Xiao-xi treatment group (western medicine group), Yiqi Jiedu Huoxue Xiaoqifang + Treatment group (Western group). After 8 weeks of continuous intervention, the urine samples of lupus mice were collected at 0, 2, 4, and 6 weeks respectively for protein quantification. After the intervention, the renal pathological changes, Hyp ) And the expression of transforming growth factor-β 1 (TGF-β 1) and connective tissue growth factor (CTGF) mRNA in renal tissue. Results: The 24 h urinary protein, Hyp content, kidney pathology, TGF-β 1 and CTGF mRNA expression in NZB / W F1 mice with lupus erythematosus were improved in different treatment groups. However, there was no significant difference between the treatment groups ; And kidney pathology showed Yiqi Jiedu Huoxue Xiao Fang Xiao Xi treatment group had the lightest degree of renal disease. Conclusion: Yiqi Jiedu Huoxue Prescription can reduce proteinuria and improve renal fibrosis in NZB / W F1 mice. The mechanism may be related to the down-regulation of TGF-β1 and CTGF mRNA expression.
