观察淀粉样β蛋白1-4 0 ( Aβ1-4 0 )对爪蟾卵母细胞表达的淋巴细胞神经递质受体 ( NR)功能的影响 ,探讨淋巴细胞 NR功能检测在阿尔茨海默痴呆( AD)病理生理学上的意义 .将提取的人外周血淋巴细胞 m RNA( 50 ng)显微注射到成熟的非洲爪蟾卵母细胞 ,在 ( 1 9.0± 1 .0 )℃条件下孵育 2 4 h后采用双电极电压钳位记录 - 60 m V维持电位时的受体激活电流 .Aβ1-4 0 于记录前 1 2 h加入孵育液 .结果发现注射人外周血淋巴细胞 m RNA的爪蟾卵母细胞能表达出乙酰胆碱 ,谷氨酸 ,多巴胺 ,5-羟色胺和 γ-氨基丁酸受体 .这些表达的受体激活电流的共性是由氯离子载流的内向电流 ,其平衡电位接近于 - 2 2 m V.Aβ1-4 0 60 nmol· L-1对卵母细胞表达的 NR功能有抑制效应 ,其 NR峰电流降低分别为乙酰胆碱 2 3% ,谷氨酸 2 7% ,多巴胺 2 5% ,维生素E有拮抗 Aβ1-4 0 效应的作用。外周血淋巴细胞 NR功能检测作为 AD外周评价指标值得更深入的研究 .
To investigate the effect of Aβ1-4 0 on lymphocyte neurotransmitter receptor (NR) expression in Xenopus oocytes and to explore the role of lymphocyte NR function in Alzheimer’s dementia AD) pathophysiological significance of extraction of human peripheral blood lymphocytes m RNA (50 ng) microinjection of mature Xenopus oocytes, (14.0 ± 1.0) ℃ under the conditions of incubation 24 h, the bipolar voltage clamp was used to record the receptor activation current at 60 mV sustaining potential.Aβ1-4 0 was added to the incubation solution 12 h before recording.Results Xenopus laevis The mother cells express acetylcholine, glutamate, dopamine, serotonin, and gamma-aminobutyric acid receptors.The commonalities of the receptor-activating currents for these expressions are inward currents carried by chloride ions with equilibrium potentials close to - 2 2 m V.Aβ1-4 0 60 nmol · L-1 had an inhibitory effect on the NR function expressed by oocytes, and the reduction of NR peak current was 23% of acetylcholine, 27% of glutamate, 25% of dopamine, , Vitamin E antagonize the effect of Aβ1-4 0 effect. Peripheral blood lymphocyte NR function test as a peripheral nerve evaluation index deserves more in-depth study.
